慢性及急性实验性自身免疫性葡萄膜炎中眼浸润细胞比较

摘要:

目的:探讨实验性自身免疫性葡萄膜炎(experimental autoimmune uveitis, EAU)眼浸润细胞在C57BL/6及B10.RⅢ品系小鼠中的差异。方法:通过完全弗氏佐剂结合光感受器间维生素A类结合蛋白在C57BL/6和B10.RⅢ品系小鼠中建立慢性及急性EAU疾病模型,通过视网膜成像技术及视觉电生理技术分析比较两种EAU小鼠临床评分,通过流式细胞技术分析疾病高峰期炎性眼组织中浸润细胞类型及比例。结果:急性EAU比慢性EAU更早发病、更早到达疾病高峰且临床评分高[1]。在炎性眼组织中,急性EAU小鼠的CD4+ T淋巴细胞和CD11b+细胞比例显著高于慢性EAU小鼠,而CD8+ T淋巴细胞、自然杀伤性细胞及自然杀伤性T细胞在两种EAU小鼠模型中无显著性差异,其中眼浸润CD11b+细胞中,急性EAU小鼠巨噬细胞及骨髓来源抑制性细胞比例显著高于慢性EAU小鼠。结论:急性EAU和慢性EAU的疾病差异与眼浸润CD4+ T淋巴细胞、巨噬细胞及骨髓来源抑制性细胞相关。

Abstract:

Objective: To compare the cellular infiltrate characters of eyes in experimental autoimmune uveitis(EAU) induced in C57 BL/6 and B10.RⅢ mice. Methods: EAU was induced by interphotoreceptor retinoid-binding protein with complete Freund's adjuvant in C57 BL/6 and B10.RⅢ mice. Clinical scores were evaluated by funduscopy and electroretinography. Cells were isolated from inflamed eyes and analyzed with flow cytometry.Results: Mice with induced acute EAU showed earlier disease onset and peak as well as higher clinical scores than chronic EAU mice. In addition to increase of CD4+ T lymphocytes, percentage of CD11 b+ cells were higher in the eyes of acute EAU mice than those in chronic EAU mice, while no differences were observed in CD8+ T cells, natural killer cells or natural killer T cells. Among those infiltrated CD11 b+ cells, percentages of macrophages and myeloid-derived suppressor cells were higher in acute EAU mice than those in chronic EAU mice. Conclusions: The difference in above mentioned aspects between acute and chronic EAU may be related with the increase of infiltrated CD4+ T lymphocytes, macrophages and myeloid-derived suppressor cells.
