复方茵陈蒿汤通过TRPV3/TLR2/NF-κB信号通路抑制痤疮丙酸杆菌诱导的皮肤炎症的机制研究

摘要:

目的:探讨复方茵陈蒿汤通过调控TRPV3/TLR2/NF-κB信号通路对痤疮丙酸杆菌诱导的巨噬细胞炎症反应的影响,阐明其治疗痤疮的作用机制。方法:采用肽聚糖刺激巨噬细胞构建痤疮丙酸杆菌感染的体外细胞模型,分别予以复方茵陈蒿汤或异维A酸干预治疗。检测各组细胞一氧化氮(NO)生成量、炎症因子(TNF-α、IL-1β、iNOS、COX2)水平及氧化应激指标(SOD、MDA、ROS);通过蛋白质印迹法(Western blot)和实时荧光定量PCR(RT-qPCR)检测TRPV3/TLR2/NF-κB通路关键分子表达。结论:与模型组比较,复方茵陈蒿汤能够显著抑制巨噬细胞中NO的生成,降低TNF-α、IL-1β、iNOS、COX2、MDA的含量,抑制ROS的荧光强度,和增加SOD的活力,差异具有统计学意义(P<0.01)。Western blot与RTqPCR结果显示,复方茵陈蒿汤还能显著下调巨噬细胞TRPV3蛋白和mRNA的表达,抑制TLR2和p-IкBα蛋白的表达,降低p-IKKβ/IKKβ和p-p65/p65的比值,上调IκBα蛋白的表达,差异具有统计学意义(P<0.01);亚细胞定位分析表明,复方茵陈蒿汤可增加细胞质p-p65蛋白表达,同时降低核内p-p65水平,差异具有统计学意义(P<0.01)。结论:复方茵陈蒿汤可通过抑制TRPV3/TLR2/NF-κB信号通路活化调控氧化应激与炎症反应,从而发挥抗痤疮作用。

Abstract:

Objective:To investigate the effect of Fufang Yinchenhao Decoction on Cutibacterium acnes(C.acnes)-induced inflammatory response in macrophages by regulating the TRPV3/TLR2/NF-κB signaling pathway,and to elucidate its mechanism in treating acne.Methods:An in vitro cell model of C.acnes infection was established by stimulating macrophages with peptidoglycan,which was then treated with Fufang Yinchenhao Decoction or isotretinoin.The levels of nitric oxide(NO),inflammatory factors(TNF-α,IL-1β,iNOS,and COX2),and oxidative stress markers(SOD,MDA,and ROS) were measured.The expression of key molecules in the TRPV3/TLR2/NF-κB pathway was detected using Western blot and RT-qPCR.Results:Compared to the model group,Fufang Yinchenhao Decoction significantly inhibited the production of NO in macrophages,reduced the levels of TNF-α,IL-1β,iNOS,COX2,and MDA,suppressed the fluorescence intensity of ROS,and enhanced the activity of SOD,with all differences being statistically significant(P<0.01).Western blot and RT-qPCR results further demonstrated that Fufang Yinchenhao Decoction significantly downregulated the expression of TRPV3 protein and mRNA,inhibited the expression of TLR2 and p-IκBα proteins,decreased the ratios of p-IKKβ/IKKβ and p-p65/p65,and upregulated the expression of IκBα protein,with all differences showing statistical significance(P<0.01).Subcellular localization analysis revealed that Fufang Yinchenhao Decoction increased the expression of cytoplasmic p-p65 while reducing the level of nuclear p-p65,with both changes being statistically significant(P<0.01).Conclusion:By inhibiting the activation of the TRPV3/TLR2/NF-κB signaling pathway and regulating oxidative stress and inflammatory responses,Fufang Yinchenhao Decoction exerts its anti-acne effects.
