目的:研究修复周围神经缺损新材料。方法:Wistar大鼠80只,切断左大腿坐骨神经,造成15 mm缺损,分别用自体神经(C组)、液氮冷冻猫胚神经(B组)、液氮冷冻猫胚神经加自体雪旺氏细胞粗制品(C组)桥接。术后4,12,24周取桥体、桥体远端坐骨神经分别行光镜观察、图像分析、电生理检测。所得数据经多因数方差分析和q检验。结果:加入自体雪旺氏细胞猫胚神经组(C组)有髓纤维数目、复合动作电位峰值恢复率、传导速度恢复率与自体神经移植组(A组)比较无显著性差异;而单纯猫胚神经组(B组)大多数指标均不及A、C两组(P<0.01)。结论:植入自体雪旺氏细胞的胚胎神经桥接周围神经缺损效果优于单纯胚胎神经,是一种良好的替代自体神经的桥接物。
Abstract:Objective: To investigate novel material to repair defect of peripheral nerve. Methods: 80 Wistar rats were contributed to the study. A 15mm segmental sciatic nerve was removed and bridged with autograft (group A)), cat fetal sciatic nerve iced in liquefy nitrogen (group B) and cat fetal sciatic nerve implanted acceptor's SC after iced in liquefy nitrogen (group C). Harvested the graft and distal regenerated sciatic nerve stump after operation 4,12,24 weeks. Microscopy, image analysis and electrophysiology methods were used the experimental. The data was analyzed with AONVA and q test. Result: There is no difference of number of myelinated axons, the recovery ratio of peak value and conduct velocity of compound action potential between group C and A, but there are significantly differences of index above between group A or C and B(P < 0.01). Conclusion: The fetal nerve implanted with acceptor SC is a ideal graft to replace autograft in peripheral nerve repair.
