俞募配穴对少弱精子症大鼠睾丸TGF-β/smads信号通路的影响

摘要:

目的:通过观察俞募脑肾配穴电针疗法对少弱精子症模型大鼠的影响,探究俞募脑肾配穴电针疗法对少弱精子症大鼠生殖功能的改善作用及对睾丸TGF-β/smads信号通路的影响。方法:40只雄性SD大鼠随机分为空白组、模型组、电针组、左卡尼汀组,每组10只。除空白组外,其余组采用腺嘌呤灌胃法制备少弱精子症大鼠模型,灌胃连续28 d,造模成功后,分别对电针组(电针额旁3线、肾俞、京门、太溪,30 min/次/d,连续28 d)、左卡尼汀组(以10 mL/kg左卡尼汀口服液灌胃,连续28 d)进行干预。治疗后取各组大鼠附睾精子检测其密度和活力率;H&E染色观察睾丸病理形态学变化;ELISA检测血清性激素睾酮、卵泡刺激素、黄体生成素、雌二醇的变化;Western blot检测TGF-β、P-smad2、P-smad3在睾丸组织中的表达水平。结果:与空白组比较,模型组大鼠睾丸组织中生精细胞变性,数量减少,排列紊乱;精子密度、精子活力率、血清睾酮水平显著降低(P<0.05),黄体生成素水平显著升高(P<0.05);与模型组比较,左卡尼汀组、电针组大鼠睾丸病理损伤显著减轻,精子密度和活力率、血清睾酮水平显著增高(P<0.05),黄体生成素水平显著降低(P<0.05)。结论:俞募脑肾配穴电针疗法可提高少弱精子症大鼠精子密度和活率,调节性激素产生,改善大鼠的生精环境,其机制可能与血清性激素水平的调控和通过TGF-β/smads信号通路,影响睾丸生精过程有关。

Abstract:

Objective: To observe the effects of Shumu Naoshen combined with acupoint electroacupuncture on oligoasthenospermia model rats, and to explore the effects of Shumu Naoshen combined with acupoint electroacupuncture on the reproductive function of oligoasthenospermia rats as well as the influence of TGF-β/smads signaling pathway on testis. Methods: A total of 40 male SD rats were randomly divided into the blank group, the model group, the electroacupuncture group, and the L-carnitine group, with 10 rats in each group. Except for the blank group, the oligoasthenospermia rat model was prepared by adenine instillation for 28 d. After a successful modeling, the electroacupuncture group(3 lines of frontal injection, Shenshu, Jingmen, and Taixi injection, 30 min·time-1·d-1, for 28 d) and the L-carnitine group(10 mL/kg L-carnitine oral infusion, for 28 d) were respectively administered. After the treatment, the sperm density and motility rate of epididymis were tested. The morphological changes of testis were observed by H&E staining. Serum sex hormones testosterone, follicle-stimulating hormone, luteinizing hormone, and estradiol were detected by ELISA. The expression levels of TGF-β, P-smad2, and P-smad3 in testicular tissues were detected by Western blot. Results: Compared to the blank group, spermatogenic cells in testis tissue of model group were degenerated, the number was decreased and the arrangement was disordered. Sperm density, sperm motility rate, and serum testosterone level were significantly decreased(P<0.05), luteinizing hormone level was significantly increased(P<0.05). Compared to the model group, the pathological damage of testicle in L-carnitine group and electroacupuncture group was significantly reduced; sperm density, motility rate, and serum testosterone level were significantly increased(P<0.05), and luteinizing hormone level was significantly decreased(P<0.05). Conclusion: Shumu Naoshen combined with acupoint electroacupuncture can increase sperm density and viability, regulate sex hormone production, and improve spermatogenesis environment of rats with oligoasthenospermia. The mechanism may be related to the regulation of serum sex hormone levels and the influence of TGF-β/smads signaling pathway on testis spermatogenesis.
