目的:构建一种基于罗丹明B为母体的新型荧光探针,考查其作为荧光探针P的光学性能,发展适合于分析检测细胞内Fe3+的可视化成像分析的荧光探针法。方法:RAW细胞中加入20μmol/LFe3+孵育30min后,再在该培养液中加入20μmol/L探针P,37℃下继续孵育30min,559nm激发,收集570~670nm波段内的荧光信号。结果:探针能够实现对细胞内Fe3+的可视化成像分析。结论:本方法探针容易制备,分析速度快,可实现结果的可视化观测。
Abstract:Objective: This paper is to construct a rhodamine-based probe P for the cell imaging detection of Fe3+.Methods: RAW cells plated on coverslips were washed with phosphate-buffered saline(PBS),followed by incubating with 20 μmol/L of FeCl3(in PBS) for 30 min at 37 °C,and then washed with PBS three times.After incubating with 20 μmol/L of P for 30 min at 37 °C,the cells were washed with PBS three times again.Results: The proposed probe P was successfully used for the cell imaging detection of Fe3+.Conclusions: As a fast and easy method,probe P can be applied to the visible recognition of Fe3+ in living cells.
