基于网络药理学探讨四妙散缓解急性痛风性关节炎的作用机制

摘要:

目的:寻找四妙散缓解急性痛风性关节炎的核心靶点及药物作用机制。方法:通过TCMSP数据库,获取四妙散全部药物的化学成分,编译Perl脚本与查询UniProt数据库,确定符合条件的化学成分对应的靶点,然后依次检索OMIM、DisGeNET、GeneCards等疾病数据库,确定急性痛风性关节炎相关的靶点。最后筛选药物与疾病的共同靶点,使用STRING数据库与Cytoscape软件,构建药物--化学成分--靶点--疾病的网络调控图,使用R语言软件对共同靶点进行筛查,寻找四妙散治疗急性痛风性关节炎的核心靶点,对核心靶点进行GO和KEGG的分析,明确四妙散治疗急性痛风性关节炎的核心靶点及作用机制。结果:四妙散的有效化学成分共64个,基因靶点197个,疾病靶点600个,共同靶点58个,预测IL-6、VEGFA、IL-1B、JUN、PTGS2、CCL2可能是四妙散治疗急性痛风性关节炎的核心靶点。GO富集分析确定了85个条目,其中生物过程主要包括细胞因子活性的调控、蛋白酶激活、核酸的表达等;KEGG通路富集分析明确了135条信号通路,其中信号通路涉及IL-17信号通路、TNF信号通路、Th17细胞分化等通路。结论:四妙散通过调控炎症的发生作用于急性痛风性关节炎,抗炎免疫效果显著,组方配伍严谨科学,值得临床推广应用。

Abstract:

Objective: To find the core targets and drug action mechanism of Si Miao decoction for the treatment of acute gouty arthritis. Methods: First of all, chemical constituents of all the drugs in Si Miao decoction were obtained through the TCMSP database. The Perl script was compiled and the UniProt database was searched to determine the corresponding targets of the chemical constituents. Then, the disease databases such as OMIM, DisGeNET, and GeneCards were retrieved in order to determine the acute gouty arthritis related targets. Finally, the common targets of the drugs and the disease were screened. STRING database and Cytoscape software were used to build a network control map of drugs-chemical constituents-targets-disease. R language software was used to screen common targets and find the core targets of Si Miao decoction for treating acute gouty arthritis. The GO and KEGG enrichment analyses were performed to clarify the core targets and mechanism of Si Miao decoction in the treatment of acute gouty arthritis. Results: There were 64 effective chemical constituents, 197 genetic targets, 600 disease targets, and 58 common targets in Si Miao decoction. It was predicted that IL-6, VEGFA, IL-1 B, JUN, PTGS2, and CCL2 might be the core targets of Si Miao decoction in the treatment of acute gout arthritis. GO enrichment analysis identified 85 entries, among which biological processes mainly included the regulation of cytokine activity, protease activation, nucleic acid expression, etc. KEGG pathway enrichment analysis identified 135 signaling pathways, of which signaling pathways involved the IL-17 signaling pathway, TNF signaling pathway, Th17 cell differentiation, etc. Conclusion: Si Miao decoction acts on acute gouty arthritis by regulating the occurrence of inflammation and has significant anti-inflammatory and immune effects. The formula of Si Miao decoction is rigorous and scientific. Hence, Si Miao decoction is worthy of clinical application.
