正交试验优选裸花紫珠水提液的壳聚糖絮凝澄清工艺

摘要:

目的:研究壳聚糖澄清剂用于裸花紫珠水提液的澄清工艺。方法:以固形物去除率和毛蕊花糖苷和异毛蕊花糖苷含量的保留率为指标,考察药液浓度、澄清剂的用量、澄清温度和搅拌速度4个因素,采用正交试验优选壳聚糖的絮凝澄清条件。结果:壳聚糖澄清剂较佳的絮凝澄清条件为水提液浓缩至0.083g/mL,壳聚糖用量为药液体积的12%(V/V),澄清温度为70℃,搅拌速度为100r/min。在此条件下固形物去除率为20.51%,毛蕊花糖苷和异毛蕊花糖苷的保留率为90.24%。结论:壳聚糖澄清剂处理裸花紫珠水提液的澄清效果良好,优选的澄清工艺稳定可行。

Abstract:

Objective:To investigate the flocculation and clarification process of the water extract of Callicarpae Nudiflorae Folium with chitosan as clarification agent.Methods:With solid removal rate and retention rate of the content of acetoside and isoacetoside as indexes,orthogonal design tests were adopted to optimize the flocculation clarification process by taking solution concentration,the amount of clarifying agent,clarifying temperature and stirring speed as factors.Results:The optimum flocculation clarification process conditions were:concentrated water extract to 0.083g/mL,the amount of chitosan 12%(V/V),the clarifying temperature 50℃and the stirring speed 100r/min.Under these conditions,the solid removal rate and retention rate of the total content of acetoside and isoacetoside were 20.51% and 90.24%,respectively.Conclusion:Clarification effect of chitosan as clarifying agent for water extract of Callicarpae Nudiflorae Folium is effective,and the optimized process is stable and feasible.
