目的:研究三叶因子3(TFF3)在人甲状腺乳头状癌组织中的表达,探讨TFF3在甲状腺乳头状癌发生、发展中的作用。方法:采用RT-PCR、免疫组织化学法检测TFF3在75例甲状腺乳头状癌和10例结节性甲状腺肿组织中的表达,并进行分析。结果:甲状腺乳头状癌组织中TFF3mRNA水平高于结节性甲状腺肿,差异有统计学意义(P<0.05);血清中TFF3差异不明显。Western blot检测结果显示,TFF3蛋白在结节性甲状腺肿基本不表达,在甲状腺乳头状癌组织高表达(P<0.05);且甲状腺癌染成棕色的区域主要位于肿瘤边缘。TFF3的表达与甲状腺乳头状癌患者的年龄、性别、肿瘤个数无关,与肿瘤直径、淋巴结转移以及TNM分期相关。结论:TFF3在甲状腺乳头状癌中表达增高,TFF3可能通过影响多种细胞因子以及炎症细胞参与甲状腺癌的发生和发展。
Abstract：Objective:To investigate the expression of TFF3in human thyroid papillary carcinoma tissues,and explore the role of TFF3in the development of papillary thyroid cancer.Methods:RT-PCR,immunohistochemistry assay were used to detect the expression of TFF3in 75cases of papillary thyroid carcinoma,and 10cases of nodular goiter tissues.Statistics methods were applied to analyze the results.Results:The level of TFF3mRNA in papillary thyroid carcinoma tissues was higher than that in nodular goiter tissues;The frequencies of TFF3in peripheral blood were no difference.TFF3protein in nodular goiter tissues was rarely seen,but expressed in papillary thyroid cancer;The dyed brown areas in thyroid cancer were mainly located on the edge of the tumor;Clinical data statistics showed that the expression of TFF3had no correlation with the patient's age,sex,tumor numbers,but related with tumor size,lymph node metastasis and TNM staging.Conclusions:The expression of TFF3increases in papillary thyroid carcinoma tissues.It may be involved in the occurrence and development of thyroid cancer by interacting a variety of cytokines and inflammatory cells.
